題名:高血壓對老化之血管內皮功能和抗氧化的影響

背景及目的:目前世界人口有老年化之趨勢,在老化人口中發現因年齡的增長罹患高血壓風險增高,且心血管功能亦隨年齡逐漸下降。有許多研究討論老化與心血管功能變化之關係,但高血壓對老化心血管功能的影響不甚清楚,故本研究欲探究高血壓對老化之血管內皮功能和抗氧化活性之影響。方法:本研究使用雄性自發性高血壓大鼠(spontaneous hypertension rat, SHR),隨機分為兩組,分別為二十四週齡大的高血壓成年組和四十八週齡大的高血壓老化組,另以同週齡之雄性正常血壓大鼠(Wistar-Kyoto rat, WKY)做為對照組。各組於實驗結束時以組織浴系統評估血管內皮功能,並進行相關血清分析,包括一氧化氮(nitric oxide, NO)和抗氧化活性(過氧化氫酶catalase, CAT、超氧歧化酶superoxide dismutase, SOD)等,最後進行各組之統計和比較。結果:本研究發現血管舒張反應方面,與同週齡WKY組相較,SHR-24組和SHR-48組二組之內皮依賴性舒張反應皆顯著下降(p < 0.05),且SHR-48組與SHR-24組相較反應則顯著減少(p < 0.05),此外,SHR-24組和SHR-48組二組之非內皮依賴性舒張反應亦顯著下降(p < 0.05)。而血清中一氧化氮含量與抗氧化酵素活性(CAT、SOD)方面,SHR-24組和SHR-48組與同週齡之WKY組比較二組含量亦顯著降低(p < 0.05),其中CAT的部分,SHR-48組顯著低於SHR-24組(p < 0.05)。結論:本研究證實高血壓會造成老化之血管舒張反應更為下降,此與一氧化氮含量和抗氧化活性(CAT, SOD)降低有關。

Backgroud and purpose: Aging has become a trend in the world. Advancing age is associated with the prevalence of hypertension and the decline of cardiovascular function. Many evidences have discussed about the relationships of aging and cardiovascular function, however, the effects of hypertension on aging-related cardiovascular function remained unclear. Therefore, the purpose of this study was to investigate the effects of hypertension on aging-related endothelial function and antioxidant activity. Methods: The male spontaneously hypertensive rats (SHR) were randomly divided into two groups:twenty-four-week-old adult SHR (SHR-24) and forty-eight-week-old aging SHR (SHR-48) groups, while the age-matched Wistar-Kyoto (WKY) rats were served as the normotensive control (WKY-24 and WKY-48) groups. At the end of experiments, the endothelial function was evaluated by the organ bath system among four groups. Furthermore, serum concentrations of nitric oxide (NO) and antioxidant activity (catalase, CAT; superoxide dismutase, SOD) were measured and compared among four groups. Results: We found that, compared with the age-matched WKY groups, the endothelium-dependent vasorelaxation was significantly (p < 0.05) impaired in SHR-24 and SHR-48 groups. This vasorelaxation was worse in the SHR-48 compared with the SHR-24 group (p < 0.05). Also, the endothelium-independent vasorelaxtion was significantly (p < 0.05) decreased in SHR-24 and SHR-48 groups. Moreover, serum concentrations of NO and antioxidant activity (CAT, SOD) were significantly (p < 0.05) reduced in both SHR-24 and SHR-48 groups, compared with their age-matched WKY groups. The CAT activity was significantly (p < 0.05) decreased in the SHR-48 group compared with the SHR-24 group. Conclusions: This study indicated that hypertension resulted in worse aging-related vasorelaxant responses. This could be associated with the decline of NO concentration and antioxidant activity (CAT, SOD).
