題名:情境害怕制約對大白鼠海馬迴細胞增生之影響:青春期前後與燈光線索性害怕制約的比較

海馬迴顆粒與錐狀細胞於個體出生後仍持續增長,但探討情境害怕制約對海馬迴細胞增生影響之相關研究以顆粒細胞為主。本研究比較情境害怕制約及燈光害怕制約對海馬迴兩種細胞增生的影響是否因青春期前後而有所差異。實驗一發現情境害怕制約於二十四小時內,能促進十週大白鼠海馬迴顆粒細胞 (F(1,22) = 706.906, p < .05) 與錐狀細胞 (F(1,22) = 540.522, p< .05) 的增生。實驗二探討情境害怕制約對海馬迴細胞增生的促進是否因青春期前後 (四至六週與八至十二週) 而有所差異。結果顯示,情境害怕制約可促進海馬迴顆粒細胞 (F(1,32) = 459.864, p < .05) 以及錐狀細胞的增生 (F(1,32) = 172.625, p < 05)。實驗三嘗試比較在情境害怕制約組 (已表現出制約行為) 與燈光害怕制約組 (無制約行為表現) 的情況下,老鼠海馬迴顆粒與錐狀細胞數目的增生情形。結果顯示,顆粒細胞數目在接受情境或燈光害怕制約後皆高於控制組 (F(1,64) = 1227.444, p < .05),情境害怕制約對顆粒細胞增生的幅度高於燈光害怕制約 (F(1,64) = 198.459, p < .05)。錐狀細胞數目在接受情境害怕制約後增生的幅度高於燈光害怕制約所促進 (F(1,64) = 57.221, p < .05)。燈光害怕制約能促進青春期後錐狀細胞增生,但不影響青春期前錐狀細胞增生 (F(1,16) = 39.416, p <. 05)。本研究之發現有助於瞭解青春期前後錐狀細胞在接受害怕制約後之增生情形。
Granular and pyramidal cells in hippocampus are continually born after postnatal period. Past research has mostly focused on the effects of contextual fear conditioning (CFC) on granular cells of hippocampal neurogenesis. The present study attempted to investigate the effects of CFC on the numbers of hippocampal granular and pyramidal neurons in rats before and after puberty, as well as a comparison of CFC and light fear conditioning (LFC) on the neurogenesis. Experiment 1 found that CFC increases the numbers of both the granular and pyramidal cells in 10 weeks-old rats. In the Experiment 2, rats before puberty (4 to 6 weeks) and after puberty (8 to 12 weeks) were undergone a CFC procedure and compared on the proliferation of their granular and pyramidal cells. Results suggested that CFC improved the proliferation of both granular and pyramidal cells. Experiment 3 investigated the effects of CFC and LFC on rats before and after puberty. Findings suggested that under these two conditionings, the number of the granular cell increased. The numbers of granular and pyramidal cells were more in CFC than in LFC. Furthermore, LFC had a significant effect on the pyramidal cells of rats after puberty. Taken together, the present study suggested that CFC increases proliferation on both the granular and pyramidal cells of rats and LFC would have effects on the pyramidal cells of rats after puberty. These results have further our understanding on the influence of fear conditioning on the hippocampal neurogenesis.
